Disregarding the commas in the given sequence, we have:

1605181612052421

Dividing the above number in groups of two digit numbers, we have:

Group 1: 16

Group 2: 05

Group 3: 18

Group 4: 16

Group 5: 12

Group 6: 05

Group 7: 24

Group 8: 21

We now replace numbers in each group with the letter in the alphabet having the requisite position. For example, the 16th letter in the alphabet is p, so the number "16" is replaced by the letter "p"

Accordingly, we have the string 'perplexu'. Clearly, we need to add the letter "s" to this string to obtain a meaningful word, that is, "perplexus".

Now, "s" is the 19th letter of the alphabet.

Consequently, the reuired next number in the given sequence is 19.
